Correct.
It supports both a DFC with a S2/M2, or a DFC3 with a Sup720.
You asked, "Will a WS-6816 work in a SUP-2U/MSFC2 without a Switch Fabric
Module?"
While the document says, "supported only with a Sup2/SFM", which is OK
because this document is centric to MSFC1/2 system.
Seperately on CCO it states this card is supported on a Sup720.
So, to answer your question, "No, it will not work on a s2/m2 without a
SFM."
