Solved
the problem was not with the mail store as mentioned by Matt (thank you)
it works just fine when configuring the HTTPS to use 443 and properly configuring PCA user name and password on unity connection .

Mail store is for connecting to Microsoft Exchange for calendar
presence integration. It is not related to Unity Connection.

Dear all
when running system troubleshooter form presence administration i have a
problem with something called mail store ( the server is unreachable) , what
is exactly the settings needed to correct this problem? , what is the port
and protocol configuration (TLS, TCP , SSL)
, I have setup the the web service protocol to HTTPS according
to Cisco documents
if you have successfully implement it please let me know any checklist for
this kind of integration
