You will not be able to hang a modem off either of the sync ports on the
2501. There are interfaces available for the 1600/3600 series, and some
of the 2500 series routers have sync/async ports on them as well, but the
2501 is definitly not one of them.
Try this URL for an example on how to get Async DDR working out of the aux
port.

I think the problem you are going to run into is the fact that the aux
port only supports speeds up to 9600 baud and there is no modem control
available to hang the modem up.
If I were you, I would trade that 2501 in for a 1601 with at least 1
WIC-1T (or better yet, 2) and set up ddr out of the serial port on it.
You certainly don't need a modem that supports v.25 bis dialing. Set the
interface for in-band dialing, and then specify a dialer string with a
keyword that matches a chat-script. Put the entire dialing string in the
chat script, you know, atdt555-1212 and all that stuff. If you are
dialing to a provider, you are also going to need to send some sort of
authentication from the router, but not receive any. Make sure you set
the chap or pap to send-only.
Here is a good page to look at for this application:

ios111/mods/3mod/3cbook/3cddr.htm
This is the DDR reference guide for IOS 11.1
